The principal research interest of the lab is to comprehensively understand the biology of specific cancer types that are common in India and/or are distinct from their western counterpart(s). To this end, we use genomic, molecular genetic, molecular pathology and cell biology tools to identify and characterize recurrent genetic alterations in tumor samples.

Positions-We are currently seeking potential PhD students and Postdoctoral candidates to work on two major projects. One involves characterisation of oncogenic function of rare p53 mutant forms by identification and characterization of their transcriptional targets as well as dissection of the mode of target gene activation at the transcription level. The work will include CRISPR-Cas9 based knock-in of p53 mutations, several cell and molecular biology based experiments in cancer cell lines and evaluation of tumorigenicity in nude mice. The other involves the study of non-canonical roles of ARID1B (one of the four alternative DNA binding subunits of the SWI/SNF chromatin remodelling complex) with a major focus on its role in functioning of enhancers and DNA repair. The work comprises proteome based screen for interactive partners, chromosome conformation capture, ChIP/RNA Seq and further characterization.

Details of current projects are given below:
  1. Characterisation of gain of function activity of non-hotspot p53 mutations identified from Indian squamous cell carcinoma patients using the CRISPR/Cas9 system.

  2. Identification and characterization of pancreatic cancer genes located within novel localized copy number alterations.

  3. Molecular Characterization of early-onset rectal cancer.

  4. Molecular analysis of genetic disorders.
